Biochem/physiol Actions
Endostatin inhibits the proliferation and migration of endothelial cells. It blocks apoptosis of cells and G1 to S phase transition in the cell cycle. It inhibits angiogenesis by blocking the pro-angiogenic activities of VEGF and FGF-basic. Mutations in this gene lead to Knobloch Syndrome (KS), a rare autosomal recessive disorder.
General description
Endostatin is a naturally occurring 20kDa polypeptide derived from the C-terminal portion of type XVIII collagen. It functions as an anti-angiogenic cytokine that is expressed in various organs with the highest levels in liver, lung and kidney. The gene encoding it is localized on human chromosome 21q22.3. Recombinant human Endostatin is a 20.2kDa protein consisting 184 amino acid residues.
Physical form
Lyophilized from 10 mM Sodium Phosphate, pH 7.5.
Reconstitution
Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/ml. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C.
