Biochem/physiol Actions
AURA/AURKA (aurora kinase A) modulates cell cycle checkpoints and plays important roles in centrosome division, cytoskeleton and cytokinesis. It sustains the integrity of chromosomes after DNA damage. The catalytic domain of AURKA possesses a serine/threonine kinase activity.
General description
AURA/AURKA (aurora kinase A) is mapped to human chromosome 20q13.2. It is also termed as STK15 (serine/threonine kinase 15). This gene codes for a protein of 403 amino acids with regulatory and catalytic domains. AURA is overexpressed in several malignancies like invasive carcinomas of ovary, head, neck and breast.
Human Aurora kinase A (GenBank Accession No. NM_003600), amino acids 2- 403 with N-terminal HIS6-tag, MW=50 kDa, expressed in a Baculovirus infected Sf9 cell expression system.
Unit Definition
One unit is defined as the amount of enzyme that will phosphorylate 1 pmol of Ser/Thr 1 peptide substarte per minute at pH 7.4 and 30°C.
