Hematopoietic progenitor kinase 1 (HPK1) acts as a tissue-specific upstream activator for the stress-activated protein kinases (SAPK)/Jun amino-terminal kinases (JNK) signaling pathway. By phosphorylating SH2 domain-containing leukocyte protein of 76 kDa (SLP-76), HPK1 effectively blocks the T cell receptor (TCR) signaling. HPK1 is implicated in the inflammatory response and in the pathogenesis of systemic lupus erythematosus (SLE). It mediates the degradation of IκB and nuclear factor-κB (NF-κB) activation by activating inhibitor of nuclear factor-κB (IκB) kinase (IKK) complex.
General description
Hematopoietic progenitor kinase 1 (HPK1) belongs to the Ste20 serine/threonine kinase superfamily. It comprises a C-terminal citron homology (CNH) domain and a total of 13 tyrosines as potential phosphorylation residues. The HPK1 gene is mapped to human chromosome 19q13.
