Application
IL-10 human has been used to study the effect of IL-10 on endothelial nitric oxide synthase and nitric oxide in African American human umbilical vein endothelial cells. It has also been used to study the effect of IL-10 on leukotriene (LT) A4 hydrolase and LTC4 synthase protein expression in and secretion from, an inflamed porcine endometrium.
Biochem/physiol Actions
IL-10 (interleukin 10) is an anti-inflammatory cytokine. IL-10 inhibits the expression of proinflammatory cytokines such as IL-1 and TNF (tumor necrosis factor)-α. Like IL-4, IL-10 enhances humoral immune responses and attenuates cell-mediated immune reactions. Polymorphisms in the IL-10 gene are associated with higher risk of preeclampsia, ankylosing spondylitis, nasopharyngeal carcinoma and basal cell carcinoma. Polymorphism in IL-10 at -1082A/G is associated with diabetic nephropathy.
General description
IL-10 (interleukin 10) is an immunosuppressive cytokine produced by a variety of mammalian cell types including macrophages, monocytes, T cells, B cells and keratinocytes. The gene is mapped to human chromosome 1q31-1q32. Recombinant human IL-10 is an 18.6kDa protein of 161 amino acid residues. Human IL-10 active on murine cells, but murine IL-10 is inactive on human cells.

Lyophilized from 5 mM Sodium Phosphate, pH 7.2.
Reconstitution
Centrifuge the vial prior to opening. Reconstitute in 5 mM Sodium Phosphate, pH 7.2 to a concentration of 0.1-1.0 mg/ml. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C.
