Mdm2 functions as an E3 ubiquitin ligase and an inhibitor of p53 transcriptional activation.
Overexpression of Murine double minute 2 (MDM2) is observed in various cancer like lung, gastric, colorectal, hepatocellular, breast, endometrial, and prostate. A common single nucleotide polymorphism is observed at 309 amino acid (from T to G) increases the susceptibility to cancer.
MDM2 is a negative regulator of the p53 tumor suppressor.
Murine double minute 2 (MDM2) is a chief regulator of the p53 tumor suppressor protein. It is a multifunctional oncoprotein that contributes to both p53-dependent and -independent roles in oncogenesis. MDM2 has various p53-independent functions that contribute to its activity as an oncogene.
General description
Murine double minute 2 (MDM2) encoding gene is localized at 12q15 in the human chromosome. It is a multidomain nuclear phosphoprotein. It possesses really interesting new gene (RING) finger domain and its function and structural integrity is required for the action of MDM2. It also has central acidic domain.
