Application
FGF-acidic from mouse has been used to induce the expression of PTPIP51 (protein tyrosine phosphatase interacting protein 51) in mouse hippocampal cells.
Biochem/physiol Actions
FGF1 (fibroblast growth factor 1) is mainly involved in cell growth, proliferation and neurogenesis. In high-fat diet (HFD) mice, FGF1 is upreglated in adipose tissue. Absence of FGF1 in mice results in aggressive diabetic phenotype as well as adipose expansion in the presence of HFD. The PPARγ (peroxisome proliferator-activated receptor γ)-FGF1 axis helps in regulating metabolic homeostasis and insulin sensitization in mice. FGF1 also participates in wound healing, post-ischemic heart repair and making of collaterals after hindlimb ischemia.
General description
FGF1 (fibroblast growth factor 1) belongs to the FGF family of genes. In humans, FGF1 is alternatively spliced into four forms: FGF1A (in kidney), FGF1B (in brain), FGF1-C and -D (in vascular smooth muscle cells and fibroblasts). FGF1 interacts with tyrosine kinase FGF receptors (FGFR1-4) with high affinity. It can translocate between the cytosol and nucleus.Recombinant murine FGF-acidic is a 15.9kDa protein consisting of 141 amino acid residues.
Physical form
Lyophilized from 5 mM Sodium Phosphate, pH 7.2 + 100 mM NaCl.
Reconstitution
Centrifuge the vial prior to opening. Reconstitute in 5mM Sodium Phosphate, pH 7.0-7.2 to a concentration of 0.5-1.0 mg/ml. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C.
