Biochem/physiol Actions
In mammary epithelial cells, protein tyrosine phosphatase, non-receptor type 6 (PTPN6) controls EGFR (epidermal growth factor receptor) signaling. It participates in the JAK/STAT (janus kinase/signal transducers and activators of transcription) signaling pathway. PTPN6 regulates cytokine receptors like interleukin- 3 (IL-3) and erythropoietin.
General description
Protein tyrosine phosphatase, full length, (Gene bank accession number NM_080549) with N-terminal GST tag, MW = 95 kDa, expressed in an E. coli expression system.
The Src homology region 2 domain-containing phosphatase-1 (SHP1)/protein tyrosine phosphatase, non-receptor type 6 (PTPN6) is a 68kDa protein present in the cytoplasm. It is mapped to human chromosome 12p13.
Physical form
Formulated in 25 mM Tris-HCl, pH 8.0, 75 mM NaCl, 0.05% Tween-20, 50% glycerol, 2 mM EDTA, 1 mM DTT, and 10 mM glutathione.
Preparation Note
Thaw on ice. Upon first thaw, briefly spin tube containing enzyme to recover full content of the tube. Aliquot enzyme into single use aliquots. Store remaining undiluted enzyme in aliquots at -70°C. Note: Enzyme is very sensitive to freeze/thaw cycles.
Unit Definition
One unit will hydrolyze 1 nmol p-nitrophenyl phosphate per minute at pH 7.4 and 30°C.
