Biochem/physiol Actions
KDM6B (lysine demethylase 6B) is responsible for the demethylation of histone H3K27. At promoters it demethylates the repressive H3K27me3 and therefore, promotes gene expression. In human and mouse fibroblasts, it demethyates the H3K27me3 at INK4a/ARF (alternate reading frame) locus, thus, promoting the expression of p16 protein, and therefore, acts as a tumor suppressor. It is also involved in inflammatory pathways in macrophages and embryonic stem cell differentiation. The expression of KDM6B is elevated in clear cell renal cell carcinoma (ccRCC) where it promotes EMT (epithelial to mesenchymal transition). Increased expression is also linked positively with lymph node metastasis, tumor size, cancer stage and poor prognosis.
General description
Human JMJD3/KDM6B histone demethylase (amino acids 1043-end), Genbank Accession No. NM_001080424, with C-terminal FLAG-tag, MW = 71 kDa, expressed in Sf9 cells via a baculovirus expression system.
KDM6B (lysine demethylase 6B)/ JMJD3 (jumonji domain-containing 3) is a major histone demethylase protein. This protein contains a JmjC domain in its C-terminal.
Physical form
Formulated in 25 mM Tris-HCl, pH 8.0, 100 mM NaCl, 0.05% Tween-20, 10% glycerol and 3 mM DTT.
Preparation Note
Thaw on ice. Upon first thaw, briefly spin tube containing enzyme to recover full content of the tube. Aliquot enzyme into single use aliquots. Store remaining undiluted enzyme in aliquots at -70°C. Note: Enzyme is very sensitive to freeze/thaw cycles.
