Biochem/physiol Actions
SET7/9 acts as a histone methyltransferase and catalyzes monomethylation of histone H3 at lysine 4 (H3K4me1) in vitro. Down-regulation of SET7/9 expression affects human embryonic stem cell differentiation. The encoded protein is implicated in the regulation of proteins and transcription factor activity. It also stimulates promoters of methylation-dependent co-recruitment through non-histone protein methylation. Mutation in the gene encoding SET7/9 is associated with serum lycopene concentrations. Elevated expression of the gene is associated with progression of hepatocellular carcinoma (HCC).
General description
SET7/9 also known as KMT7/SET domain containing lysine methyltransferase 7 (SETD7), is encoded by the gene mapped to human chromosome 4q31.1. The encoded protein belongs to SET domain-containing protein family.
Human recombinant SET7/9, amino acids 52-end, with N-terminal GST tag, GenBank Accession No. NM_030648, expressed in a E. coli expression system. MW = 61 kDa,
Physical form
Formulated in 25 mM Tris-HCl, pH 8.0, 100 mM NaCl, 0.05% Tween-20, 30% glycerol and 3 mM DTT.
Preparation Note
Thaw on ice. Upon first thaw, briefly spin tube containing enzyme to recover full content of the tube. Aliquot enzyme into single use aliquots. Store remaining undiluted enzyme in aliquots at -70°C. Note: Enzyme is very sensitive to freeze/thaw cycles.
Unit Definition
One unit is defined as the amount of enzyme required to methylate 1 pmol of substrate/min at 37°C.
