Phosphodiesterases (PDEs) function as regulators of cyclic nucleotide levels. PDE11A is a dual-specificity PDE that might be involved in controlling both cGMP and cAMP levels under physiological conditions. A genome-wide study shows that the inactivation of this gene is linked with adrenocortical hyperplasia. Polymorphisms in this gene are also linked with ACTH (adrenocorticotropic hormone)-independent macronodular adrenal hyperplasia (AIMAH). PDE11A is also involved in the control of cortisol secretion. In Carney complex (CNC) patients with germline PRKAR1A mutations, polymorphisms in PDE11A might influence the development of testicular and adrenal tumors by acting as a modifier of the phenotype.
General description
PDE11A (phosphodiesterase 11A) belongs to the PDE protein family, and is expressed in three isoforms. Its mRNA has been found to be expressed in skeletal muscle, prostate, kidney, liver, pituitary, and salivary glands and testis. All PDEs contain a conserved PDE catalytic domain at the C-terminal, which also contains the PDE signature motif HDX2HX4N. This gene is localized to human chromosome 2q31–2q35.Human PDE11A4 (GenBank Accession No. BAB62712), full length with N-terminal GST tag, MW = 131kDa, expressed in a Baculovirus infected Sf9 cell expression system.
Physical form
Formulated in 25 mM Tris-HCl, pH 8.0, 100 mM NaCl, 0.05% Tween-20, 50% glycerol, 5 mM glutathione and 3 mM DTT.
Preparation Note
Thaw on ice. Upon first thaw, briefly spin tube containing enzyme to recover full content of the tube. Aliquot enzyme into single use aliquots. Store remaining undiluted enzyme in aliquots at -70°C. Note: Enzyme is very sensitive to freeze/thaw cycles.
