Biochem/physiol Actions

HDAC-11 (histone deacetylase 11) is responsible for deacetylation of nuclear histone and nonhistone proteins, including transcription factors. HDAC-11 catalyzes the deacetylation of histone H4. It binds to CDT1 (DNA replication factor) and is present with S phase-associated replication origins. HDAC-11 is upregulated in various carcinomas. Absence of it results in cell death and suppresses metabolic activity in colon, prostate, breast and ovarian cancer cell lines.

General description

HDAC-11 (histone deacetylase 11) is a zinc-dependent HDAC. It belongs to the HDAC family of proteins and HDAC class IV. It is highly conserved among various species. The mRNA is present in the brain, skeletal muscle, kidney and testis. The gene is mapped to human chromosome 3p25.Human HDAC11, full length, GenBank Accession No. BC009676, with N-terminal GST tag, MW = 66kDa, expressed in baculovirus expression system.

Physical form

Formulated in 25 mM Tris-HCl, pH 8.0, 100 mM NaCl, 0.05% Tween-20 and 50% glycerol.

Preparation Note

Thaw on ice. Upon first thaw, briefly spin tube containing enzyme to recover full content of the tube. Aliquot enzyme into single use aliquots. Store remaining undiluted enzyme in aliquots at -70°C. Note: Enzyme is very sensitive to freeze/thaw cycles.

Unit Definition

One unit is defined as the amount of enzyme required to deacetylate 1 pmol of substrate/min at 37°C.
