General description

Chemotaxis, or directed cell movement is a fundamental cellular process, necessary for embryonic development, angiogenesis, wound healing, and immune responses, as well as pathological events associated with inflammation and cancer metastasis [Lauffenburger & Horwitz, 1996]. Prior to cell body translocation or chemotaxis, a cell must first extend a dominant pseudopodium (lamellipodium) in the direction of the chemoattractant gradient [Lauffenburger & Horwitz, 1996; Parent & Devreotes, 1999]. Once a dominant pseudopodium is formed, cell movement can begin in the direction of the gradient. Formation of a directional pseudopodium is tightly controlled by signal transduction mechanisms that temporally and spatially regulate changes in focal adhesions, microtubules, and the actin/myosin cytoskeleton [Ridley et al., 1992; Schmidt et al., 1993; Manes et al., 1999; Nobes & Hall, 1999; Eddy et al., 2000].

The CHEMICON Pseudopodia Purification Kit is a novel system designed to specifically isolate and purify extending pseudopodia proteins from the cell body [Cho & Klemke, 2002]. This Kits provides a quick and efficient system to purify pseudopodia proteins and their responses to various natural or synthetic compounds that specifically impact the microenvironment of the pseudopodial organelle including signaling processes, integrins or other adhesion receptors, and unique gene products. This kit provides a novel method for pseudopodia protein purification; identify signaling complex formation as a result of chemotaxis stimuli.
