General description
Human native nucleosomes purified from HeLa cell nuclei, digested with micrococcal nuclease, and purified by glycerol gradient and ion exchange chromatography.
Nucleosomes are the basic units of chromatin, and are comprised of approximately 147 bp of DNA spooled around a central octamer of core histones, H2A, H2B, H3, and H4. The arrangement of nucleosomes in chromatin can be altered by post-translational modifications (PTM) occurring on the C- and N-terminal tails of the core histones. Histone modifications can either provide or hinder access of key transcriptional complexes to DNA sequences and can regulate the expression of genes. These effects can be deciphered by applications such as chromatin immunoprecipiation (ChIP), which requires high quality chromatin, and essential tools such as histone proteins and appropriate enzymes, to assemble chromatin in vitro.
