General description

Tris HCl, or Trizma® hydrochloride, is a widely utilized acidic buffer in the physiological pH range of 7.3 to 7.5. With a pKa of 8.08 at 25°C, it offers a pH range of 7.0 to 9.0, aligning with living organisms′ typical pH. Essential in various biological, cell biology, and biochemistry applications, Tris HCl is crucial for effective pH control in buffer solutions. Notably, Tris base and Tris hydrochloride alone lack sufficient buffering capacity, prompting their combination within the pH range of 7 to 9.

Tris buffers play a key role in techniques like protein electrophoresis and western blotting, serving in low ionic strength buffers for protein transfer. They are also integral to SDS-PAGE gels, running buffers, and blotting buffers. Additionally, Tris salts, especially in TAE and TBE buffers, are extensively employed in DNA agarose electrophoresis. These buffers are pivotal in diverse applications, including biochemistry, protein purification, and electrophoresis, showcasing the versatility and importance of Tris HCl in maintaining optimal pH for biochemical and cell biology research.

Applications

Trizma® hydrochloride has been used as a buffer solution for staining procedure, reduction and alkylation studies and in situ hybridisation procedure.

Features and benefits

  • Ideal as a Biological buffer for Cell Biology and Biochemical research
  • Effective Buffering from pH 7-9 with a pKa of 8.1 (25 °C)
  • Tested to confirm low levels of heavy metal contamination, ensuring suitability for various applications

Other notes

The pH values of all buffers are temperature and concentration dependent. For Tris buffers, pH increases about 0.03 unit per °C decrease in temperature, and decreases 0.03 - 0.05 unit per ten-fold dilution. For precise applications, use a carefully calibrated pH meter with a glass/calomel combination electrode.
